The Uses of Benzydamine hydrochloride

Benzydamine hydrochloride is an anti-in-flammatory drug used extensively in Europe and recently introduced in Canada to help re duce painful mucositis. Benzydamine provides short-term topical anesthesia and extended pain relief apparently through its anti-inflammatory properties. Studies have shown it to be effective in a range of painful conditions, including aphthous ulceration, chemotherapy-induced mucositis, and radiation-induced mucositis.

Manufacturing Process

To a solution of 175 g of anthranilic acid methyl ester in 2 L of water and 120 ml of concentrated hydrochloric acid at 25°C was added concentrated solution of 80 g sodium nitrite. The product was dissolved in solution of 500 g NaOH in 1.5 L of water. To this solution under nitrogen was added 400 g of sodium bisulfite. The mixture was stirred for 6 hours at 75°C under nitrogen. The obtained solid product was dissolved in water and then to the solution was added 750 ml glacial acetic acid. The yield of 1-benzyl-3-(3-(dimethylamino) propoxy)-1H-indazole 70%, M.P. 154-156°C.
In practice it is usually used as monohydrochloride salt.

It is a nonsteroidal anti-inflammatory drug used both topically and systemically. It has been reported as a sensitizer and a photosensitizer.

Side Effects

The major side effects of benzydamine hydrochloride are oral numbness (reported prevalence: 10 percent), and oral burning/stinging (reported prevalence: 8 percent). Oral discomfort appears to be more of a problem in patients with extensive ulceration/infammation. The oral discomfort can sometimes be improved by diluting the benzydamine hydrochloride with an equal volume of water.It is thought that benzydamine hydrochloride does not precipitate peptic ulceration.

Safety Profile

Poison by intraperitoneal, subcutaneous, and intravenous routes. Moderately toxic by ingestion. An experimental teratogen. Other experimental animal reproductive effects. An eye irritant. A nonsteroidal antiinflammatory analgesic. When heated to decomposition it emits very toxic fumes of HCl and NOx.
