The Uses of ATROPINE METHYL NITRATE
Atropine methyl nitrate has been used to suppress mucus production and respiratory secretions in rats. It has also been used to study its effects on xanomeline-induced suppression of serum tumor necrosis factor.

Manufacturing Process
There are 2 methods of synthesis of methyl atropine nitrate:
1) To a solution of 28.9 g atropine of in 100 g of methanol was added at
110°C for 2 hours 7.7 g methyl nitrate. Methanol was evaporated and methyl
atropine nitrate was crystallysed, M.P. 163°C.
2) To a solution of 70.4 g methyl atropine sulfate (prepared from methyl
atropine chloride and silver nitrate) in water was added aqueous solution of
26.1 g barium nitrate. Barium sulfate was deleted by filtration. Filtrate was
concentrated and the methyl atropine nitrate was obtained.

Biochem/physiol Actions
Atropine methyl nitrate is a muscarinic acetylcholine receptor antagonist that does not cross the blood brain barrier. Atropine methyl nitrate has been used for its peripheral muscarinic effects (bladder, respiratory, to block parasympathetic signaling to the heart, etc.) and to separate central from peripheral effects or protect against peripheral side effects when using muscarinics that do cross the blood brain barrier.
Safety Profile
Poison by intraperitoneal, intravenous, and subcutaneous routes. Moderately toxic by ingestion. Human systemic effects: mydriasis. Experimental reproductive effects. When heated to decomposition it emits toxic fumes of NOx. See also ATROPINE and NITRATES.
