The Uses of Methyl Green

Methyl Green powder dye is used in various staining methods in microscopy. It is also used with Pyronine Y dye for one stage DNA (green) and RNA (red) staining method. It is also used as a counterstain with other reactions, such as demonstration of enzymatic activity and for mitochondrial staining according to Altmann. Methyl green solution is a suitable counterstain for chloroacetate esterase, nonspecific esterase, alkaline phosphatase, peroxidase, naphthol AS acetate esterase, and acid phosphatase.

Definition

ChEBI: Methylene green is an organic chloride salt having 3,7-bis(dimethylamino)-4-nitrophenothiazin-5-ium as the counterion. It stains nuclei green and is sometimes used as a counterstain to red or purple primary stains. It has a role as a fluorochrome and a histological dye.
