Dabigatrul is another new oral anticoagulant approved by the US Food and Drug Administration after warfarin. It is a non-peptide thrombin inhibitor, and plays an anticoagulant effect by specifically and selectively blocking thrombin (free type or binding type) activity. It has the characteristics of oral administration, strong efficacy, no special medication monitoring, and less drug interaction. It is a major progress in the field of anticoagulant therapy and the prevention of potentially fatal thrombus, and has a milestone significance. Orally absorbed by gastrointestinal tract, dabigatrun is transformed in vivo into dabigatrun with direct anticoagulant activity. By binding to the fibrin-specific binding site of thrombin, dabigatrun prevents fibrinogen from splitting into fibrin, thus blocking the final step of coagulation waterfall network and thrombosis. Dabigatran can also be dissociated from fibrin-thrombin complex to exert reversible anticoagulant effect.
