Exemestane is an irreversible steroid aromatase inhibitor with a structure similar to its natural androstenedione substrate. Estrogen deprivation by aromatase inhibition is an effective and alternative treatment for hormone-dependent breast cancer in postmenopausal women with advanced breast cancer that has not progressed naturally or artificially after tamoxifen therapy.

Uses and Applications:

Exemestane is a steroid aromatase inactivating agent. Its structure is similar to that of androstenedione, the natural substrate of aromatase, and it is a pseudo-substrate of aromatase. Since estrogen in postmenopausal women is mainly produced by androgen (produced by adrenal cortex) in the peripheral tissue under the action of aromatase transformation, this product by irreversibly binding with aromatase active site to inactivate it, thus significantly reducing the blood circulation of estrogen in menopausal women. This product has no significant effect on the biosynthesis of adrenocorticosteroid, even when the concentration is higher than aromatase inhibition concentration 600 times, it has no significant effect on other enzymes in the corticosteroid production pathway.
