Pharmacology
Tianeptin sodium is a tricyclic antidepressant, and its antidepressant mechanism is different from that of traditional tricyclic antidepressants. It can increase the reuptake of 5-HT in synaptic cleft, and may have the effect of improving 5-HT neuronal conduction.No affinity for choline or adrenaline receptors.Antidepressant efficacy was similar to tricyclic, but tolerability was superior to tricyclic.In animal experiments, it can increase the spontaneous activity of pyramidal cells in hippocampus and accelerate the recovery after its function is inhibited.Chemicalbook increases the reabsorption of serotonin by neurons in the cerebral cortex and hippocampus.The absorption of tianepudine sodium in the digestive tract is rapid and complete.The distribution is rapid, with a PPB of 94%.The liver was metabolized completely by β-oxidation and N-demethylation. The clearance terminal was short and 2.5h, and a very small amount of the original drug (8%) was excreted from the kidney, mainly its metabolites were excreted from the urine.Clearance of t1/2 was prolonged for 1h in patients with renal insufficiency.
